Rate-limiting Description

"5-lipoxygenase (5-LO) is the rate-limiting enzyme in the synthetic pathway for cysteinyl leukotrienes (CysLTs), bronchoconstricting agents that are overproduced in asthma." (16415251)

"These results were explained after a comparison of the rates of synthesis of these 5-lipoxygenase metabolites in the presence and absence of added arachidonic acid which led to the conclusion that leukotriene A hydrolase, the enzyme catalysing the formation of LTB4, was saturated with substrate and rate-limiting for LTB4 synthesis during A23187 stimulation." (1311589)

"Therefore, the present study determined the abundance of the mRNA for arachidonate 5-lipoxygenase (5-LO; arachidonate:oxygen 5-oxidoreductase, EC 1.13.11.34), which is the rate-limiting enzyme in leukotriene synthesis, in a series of human brain tumors." (1357659)

Regulatory Information

Regulatory type

Detail

key enzyme;

"We now demonstrate that celecoxib inhibits 5-lipoxygenase (5-LO), a key enzyme in leukotriene (LT) biosynthesis." (18692027)

phosphorylation;MAPK-activated protein kinase 2

"promotion of phosphorylation at Ser-271 by MAPK-activated protein kinase 2 and arachidonic acid" (11844797)

phosphorylation;PKA

"PKA phosphorylates 5-LO on Ser-523, which inhibits the catalytic activity of 5-LO and reduces cellular LT generation" (15280375)

transcriptional factor;Sp1(6667)

"the GC-rich part of the 5-lipoxygenase gene promoter, including a novel Sp1 site, appear important for basal (rather than upregulated) transcription of 5-lipoxygenase gene in monocytic cells" (16413224)

transcriptional factor;TGFbeta(7040)

"Sequence analysis and deletion studies indicate the existence of up to four Smad binding elements and at least one TGFbeta responsive element far downstream of the transcriptional start site of the 5-lipoxygenase gene." (16919603)

others;"ANXA7(310),p53(7157)"

"ANXA7 and p53 can distinctly regulate LOX transcription that is potentially relevant to the arachidonic acid -mediated cell growth control in tumor suppression." (17018618)
