Pathway from KEGG

Amino Acid Metabolism; Glutamate metabolism; map00251

Nucleotide Metabolism; Purine metabolism; map00230

Human (9606)

Genome localization

4q12[5471 ],

Rate-limiting Description

"The reciprocal plots for 5-phosphoribosyl-1-pyrophosphate (PRPP) of liver and hepatoma enzymes gave apparent KmS of 2 microM for adenine phosphoribosyltransferase and 4 microM for HGPRT, showing two orders of magnitude higher affinities for PRPP than that of the rate-limiting enzyme of de novo purine synthesis, amidophosphoribosyltransferase (EC (Km = 400 to 900 microM)." Regulatory Information

Upstream transcription factor

Regulatory type

regulatory enzyme;

"De novo synthesis of purines at the regulatory enzyme sites of PRPP amidotransferase and IMP dehydrogenase were the major targets of the agent." (7756677)

transcriptional factor;"NRF-1(4899),Sp1(6667)"

"The human GPAT and AIRC genes are divergently transcribed from a 558 bp intergenic promoter region. Cis-acting sites and transcription factors important for bidirectional expression were identified. A cluster of sites between nt 215 and 260 are essential, although not sufficient, for expression of both genes. Two proteins from HepG2 cell nuclear extract, identified as NRF-1 and Sp1, bound to the promoter at sites within the 215-260 region." (9108165)
